FOXO4 (FOXO4-DRI) is a D-amino acid retro-inverso peptide engineered to replicate the FOXO4 interaction domain that sequesters p53 within senescent cells. Competitive displacement of the endogenous FOXO4-p53 complex selectively triggers apoptosis in p21-high senescent cells in preclinical culture models. The retro-inverso design confers protease resistance; selectivity for senescent over proliferating cells is the mechanistic specification driving its use in cellular senescence research.
